MICROBIOLOGY: PART 4 (STUDENT PROJECT)


4.0 Introduction
Water microbiology is concerned with the microorganisms that live in water, or can be transported from one habitat to another by water. Water can support the growth of many types of microorganisms. This can be advantageous. For example, the chemical activities of certain strains of yeasts provide us with beer and bread. As well, the growth of some bacteria in contaminated water can help digest the poisons from the water. Because of that, we have decided to take a few samples of water source to investigate the quantity of microorganism in each of these water sample.


4.1 Objectives
1.       To investigate the presence of microorganism in water
2.       To determine which of those water samples contain the most and the least microorganism
3.       To know which one of those water samples is the safest to be consumed


4.2 Materials and Apparatus
1.                   Water sample from the drain, distilled water, purified water and boiled water.
2.                   Micropipette
3.                   4 universal bottles
4.                   4 agar plates
5.                   Hockey stick (cell spreader)
6.                   Incubator
7.                   Parafilm


4.2 Methodology
1.       Autoclave all the apparatus that we will use.
2.       Take 10ml of sample water from the drain using a micropipette.
3.       Put the sample water inside the universal bottle. (Make sure you close the lid as soon as you transfer the water in it)
4.       Prepare an agar plate.
5.       Transfer the sample water on the agar plate.
6.       Spread the water on the agar using hockey stick (cell spreader) that has been sterilize.
7.       Seal the plate with parafilm.
8.       Repeat step 2 till 7 using distilled water, purified water and boiled water.
9.       Invert the plates and incubate at 37oC for 24 hour





4.3 Results




4.4 Discussion
What we have discussed is there are presence of microorganism in the sample water that we have investigated. The drain water sample contained the biggest amount of microorganism while purified water contained the smallest amount of microorganism after 24 hours. The safest sample water to be consumed is the purified water while the drain water is extremely dangerous to be consumed.


4.5 Conclusion
       What we can conclude from this experiment is water is one of those mediums that microorganism live in. We also concluded that we should drink purified water because it’s the safest source of water. Second safest water source is distilled water and then boiled water. And the most dangerous source of water is drain water.So we should not consumed the drain water because it might gives us dangerous diseases caused by bacterias.
